Quadruplexes. What else ?

Quadruplexes may find applications in areas ranging from nanotechnology (as nanodevices), biotechnology, to medicinal chemistry. The 3’ G-rich telomeric overhang may adopt a G-quadruplex structure. We have identified several series of quadruplex ligands. Quadruplex ligands not only bind to preformed quadruplexes, but increase the association rate of these structures, acting as G4 chaperone agents. An enzymatic assay revealed that some, but not all, G4 ligands may affect the processivity of telomerase. However, their effects on telomerase are overestimated in the classical “TRAP” assay. Furthermore, these compounds may also target non telomeric quadruplexes. We are now exploring the sequence repertoire of intramolecular quadruplexes by comparing a variety of sequences.
