N7-methylguanosine (m7G) modification of tRNAs is crucial for proper folding and stability of the cellular tRNA pool. It is catalyzed by METTL1 , a methyltransferase overexpressed in a large proportion of malignancies and a promising target for therapeutic purposes. Despite its critical role in cell proliferation, there is a substantial number of cancers insensitive to METTL1 inhibition and its genetic abrogation. Helena will present her progress towards the characterization of molecular pathways underlaying METTL1 resistance in human cancer cells.
