"Building an integrated causal model for conserved eukaryotic processes"

The seven-transmembrane signalling protein family of G protein-coupled receptors (GPCRs) mediate cellular responses to external stimuli and consequently represent significant targets for therapeutics. These receptors can respond to many types of ligands and couple to several different intracellular binding partners, which results in highly variable and tuneable downstream responses. The Nietlispach group is interested in the role of conformational dynamics in activation at these receptors and how the multifaceted characteristics of GPCR signalling is encoded in the energy landscapes of these highly dynamic proteins. We will discuss how a variety of factors including drug binding, lipid interactions, and mutations impact the equilibrium of conformations occupied by GPC Rs and the role of Nuclear Magnetic Resonance (NMR) spectroscopy in characterising transient solution conformations. We will also demonstrate how a range of complementary techniques, including pharmacological assays, molecular dynamics simulations, and biophysical assays, aid in understanding the effects of receptor dynamics on downstream intracellular signalling.
