The linear sequence property of genetic information provides critical prior knowledge when interpreting density maps of DNA , RNA, or amino acid chains. It follows that there are persistent gaps in our understanding of biological materials that, like the organization of lipids in a fluid bilayer, lack primary sequence information. I will discuss our efforts to understand the membrane-associated synthesis of chlorophyll, including initial efforts to falsify molecular simulations using halogenated lipid derivatives as contrast agents for cryo-EM imaging.
