DESCRIPTION:We have recently identified a feedback network between two pho
 sphatases (PP1 and PP2A-B56) and two kinases (Aurora B and Mps1) that ensu
 res a rapidly switchable spindle checkpoint response at kinetochores. I wi
 ll present this data and use it as a paradigm to highlight why investigati
 ng kinase-phosphatase coupling is important if we are to properly understa
 nd signal responses. Although kinases and phosphatases are antagonistic fo
 rces that turn signals on or off\, they also work together in many differe
 nt contexts to shape the right type of response. I will highlight some exa
 mples of this and present a system we have established to investigate this
  further\, which allows the dynamic control of endogenous phosphatase comp
 lexes within defined subcellular compartments in mammalian cells.
