DESCRIPTION:Along with many other scientific fields\, psychiatry - and neu
roscience more\ngenerally - has been transformed by the digital revolution
. Brain imaging\nnow has a central role in the development of our understa
nding in\npsychophysiological processes that are widely accepted as formin
g the\nbiological substrate for clinical phenotypes. Major Depressive Diso
rder\n(MDD) in adult populations is now well characterised in terms of bra
in\nstructure and function from quantitative meta-analyses of the extant\n
literature\, with varying degrees of support for current neurobiological\n
models.\n\nMR-IMPACT is a Medical Research Council funded adjunctive study
to the\nIMPACT (improving mood with psychoanalytic therapy or cognitive b
 ehaviour\ntherapy) clinical trial. Whilst post-treatment follow-up assessm
ents are\nongoing\, baseline (pre-treatment) data are available for 128 pa
tients (aged\n12-17 years) and 40 matched control participants. Assessment
s include\nmagnetic resonance imaging (MRI) of anatomy\, functional MRI du
ring tasks\nsensitive to affective bias in memory\, response inhibition an
d facial\nprocessing as well as during task-absent (i.e. rest) conditions.
\n\nPreliminary results indicate some main effects of brain connectivity\,
but\nalso that the trajectories during adolescence of brain structure and
\nfunction\, as well as cognition\, are altered in MDD. Bearing in mind th
e \nstrong prior literature on the importance of early years experiences t
o the\nprevalence of MDD in later life\, and the well established brain ch
anges\nassociated with MDD in adulthood\, the emergence of depressive symp
toms\nduring the second decade of life appears to be mediated by altered\n
trajectories off brain maturation kindled by pre-existing vulnerabilities.
