DESCRIPTION:Small RNAs are able to guide DNA methylation at ta
 rget loci through Argonaute-mediated recruitment o
 f chromatin remodelling factors.  DNA methylation 
 can be actively removed by DNA glycosylases\, whic
 h target specific regions of the genome for demeth
 ylation.  However\, it is unknown what factors pro
 vide targeting specificity for DNA glycosylases. I
 t has been hypothesized that small RNAs could prov
 ide the required specificity.\n\nI am currently in
 vestigating a locus which is targeted for demethyl
 ation by DNA glycosylases and also requires Argona
 utes in order to remain demethylated.  Demethylati
 on at this locus is also required for the regulati
 on of an adjacent gene which may itself be involve
 d in DNA demethylation itself.\n\nIn the future\, 
 I will directly test the hypothesis that small RNA
 s can function through Argonaute proteins in order
  to provide a specificity factor for directing DNA
  demethylation. This research will improve our und
 erstanding of how genes are targeted for DNA demet
 hylation in plants as well as insight into the int
 eraction between RNA silencing and DNA demethylati
 on.
