DESCRIPTION:The activation of the zygotic genome (ZGA) marks t
he first major wave of transcription in the develo
ping embryo and is crucial for developmental progr
ession. To identify the molecular details of this
process\, we use stem cells to model ZGA in vitro.
Using novel single cell sequencing technologies i
n naïve embryonic stem cells\, we identified human
8-cell like cells (8CLCs)\, that resemble the hum
an 8-cell embryo at the time of zygotic genome act
ivation. 8CLCs represent a unique novel model syst
em to study processes regulating human ZGA in vitr
o and will provide critical insights into early ev
ents in human embryogenesis in vivo.
